Nuclear Roadblocks for mRNA Export

چکیده
The THO complex and Sub2 RNA helicase have been shown to function in both transcription and mRNA processing. Rougemaille et al. (2008) now uncover evidence that THO/Sub2 coordinates mRNA processing and nuclear export.

An Interaction between KSHV ORF57 and UIF Provides mRNA-Adaptor Redundancy in Herpesvirus Intronless mRNA Export
The hTREX complex mediates cellular bulk mRNA nuclear export by recruiting the nuclear export factor, TAP, via a direct interaction with the export adaptor, Aly. Intriguingly however, depletion of Aly only leads to a modest reduction in cellular mRNA nuclear export, suggesting the existence of additional mRNA nuclear export adaptor proteins. In order to efficiently export Kaposi's sarcoma-assoc...
متن کاملInner nuclear envelope protein SUN1 plays a prominent role in mammalian mRNA export
Nuclear export of messenger ribonucleoproteins (mRNPs) through the nuclear pore complex (NPC) can be roughly classified into two forms: bulk and specific export, involving an nuclear RNA export factor 1 (NXF1)-dependent pathway and chromosome region maintenance 1 (CRM1)-dependent pathway, respectively. SUN proteins constitute the inner nuclear envelope component of the l I: nker of N: ucleoskel...
متن کاملThe N-terminal domain of Nup159 forms a beta-propeller that functions in mRNA export by tethering the helicase Dbp5 to the nuclear pore.
Nuclear export of mRNA in eukaryotic cells is mediated by soluble transport factors and components of the nuclear pore complex (NPC). The cytoplasmically oriented nuclear pore protein Nup159 plays a critical role in mRNA export through its conserved N-terminal domain (NTD). Here, we report the crystal structure of the Nup159 NTD, refined to 2.5 A. The structure reveals an unusually asymmetric s...
متن کاملConnections between the processing and nuclear export of mRNA: evidence for an export license?
A lthough the mechanisms regulating the nuclear export of eukaryotic mRNAs remain poorly understood, it has been known for some time that premRNAs, i.e., mRNAs that retain functional splice sites, are actively retained in the nucleus by what are termed splicing commitment factors (1, 2). The subsequent nuclear export of mature mRNAs therefore could be perceived as being largely caused by the re...
متن کاملHsp16p is required for thermotolerance in nuclear mRNA export in fission yeast Schizosaccharomyces pombe.
Export of mRNA from the nucleus to the cytoplasm is one of the essential steps for eukaryotic gene expression. In the fission yeast Schizosaccharomyces pombe, heat shock stress at 42 degrees C causes block of mRNA export from the nucleus. We now report that saline and ethanol stresses also inhibit nuclear mRNA export, resulting in accumulation of bulk poly (A)+ RNA, as well as a specific mRNA, ...
